Ribosomal Subunit Exchange in Dictyostelium Purpureum
نویسندگان
چکیده
The rapid and frequent exchange of the 70S ribosomes in Escherichia coli, with their 50S and 30S subunits, has been established by kinetic studies and heavy isotope techniques (1-4) . While these studies were in progress, it was reported that ribosomes of Candida krusei, a eucaryote, undergo frequent exchange of ribosomal subunits (5) . It has been suggested that ribosomes dissociate and re-form between successive rounds of protein synthesis . The mechanism of polypeptide initiation in procaryotes has been well documented (6-8) . On the other hand, still very little is known about protein initiation in eucaryotic organisms . Early works on ribosomal synthesis in HeLa cells suggested that ribosomal subunits participate in polyribosome formation prior to being assembled in single monosomes (9, 10). It has also been shown that vaccinia-virus RNA becomes associated with the smaller ribosomal subunit before appearing on polysomes (11) . The cellular slime molds have been mainly studied as a model organism for problems concerning morphogenesis and differentiation (12, 13) . The life cycle can be easily subdivided into a phase of growth (cell division) and differentiation . During the growth phase, the amebas feed on bacteria and divide by mitosis approximately every 3 hr. These organisms thus offer great advantages to study ribosome dissociation and re-assembly by both kinetics and heavy isotope techniques .

Identification and characterization of a Dictyostelium discoideum ribosomal protein gene.
We have identified a developmentally repressed large-subunit ribosomal protein gene of Dictyostelium discoideum based on sequence similarity to other ribosomal proteins. Protein rpl7 is homologous to large subunit ribosomal proteins from the rat and possibly to Mycoplasma capricolum and Escherichia coli, but is not similar to three sequenced ribosomal proteins in Dictyostelium. The rpl7 gene is...
متن کاملSecondary structure of the Dictyostelium discoideum small subunit ribosomal RNA.
We have used comparative analyses of prokaryotic and eukaryotic small subunit ribosomal RNAs to deduce a secondary structure for the Dictyostelium discoideum 18S rRNA. Most of the duplex regions are evolutionarily conserved in all organisms. We have taken advantage of the variation to the D. discoideum sequence (relative to the yeast and frog 19S rRNAs) to identify additional helical regions wh...
متن کاملrRNA maturation as a "quality" control step in ribosomal subunit assembly in Dictyostelium discoideum.
In Dictyostelium discoideum, newly assembled ribosomal subunits enter polyribosomes while they still contain immature rRNA. rRNA maturation requires the engagement of the subunits in protein synthesis and leads to stabilization of their structure. Maturation of pre-17 S rRNA occurs only after the newly formed 40 S ribosomal particle has entered an 80 S ribosome and participated at least in the ...
متن کاملSerological Investigations of Cell Adhesion in the Slime Molds, Dictyostelium Discoideum, Dictyostelium Purpureum, and Polysphondylium Violaceum
1. Antibodies to slime molds were produced by injecting D. discoideum and D. purpureum amebas from 48 hour cultures into rabbits. 2. Anti-D. discoideum and anti-D. purpureum sera caused agglutination of homologous amebas from 24 to 26 hour cultures, agglutination of certain heterologous amebas from 30 to 36 hour cultures, and agglutination of all heterologous amebas from 43 to 48 hour cultures....
متن کاملdictyBase 2013: integrating multiple Dictyostelid species
dictyBase (http://dictybase.org) is the model organism database for the social amoeba Dictyostelium discoideum. This contribution provides an update on dictyBase that has been previously presented. During the past 3 years, dictyBase has taken significant strides toward becoming a genome portal for the whole Amoebozoa clade. In its latest release, dictyBase has scaled up to host multiple Dictyos...
